    Mapping Myocardial Elasticity with Intracardiac Acoustic Radiation Force Impulse Methods

    <p>Implemented on an intracardiac echocardiography transducer, acoustic radiation force methods may provide a useful means of characterizing the heart's elastic properties. Elasticity imaging may be of benefit for diagnosis and characterization of infarction and heart failure, as well as for guidance of ablation therapy for the treatment of arrhythmias. This thesis tests the hypothesis that with appropriately designed imaging sequences, intracardiac acoustic radiation force impulse (ARFI) imaging and shear wave elasticity imaging (SWEI) are viable tools for quantification of myocardial elasticity, both temporally and spatially. Multiple track location SWEI (MTL-SWEI) is used to show that, in healthy in vivo porcine ventricles, shear wave speeds follow the elasticity changes with contraction and relaxation of the myocardium, varying between 0.9 and 2.2 m/s in diastole and 2.6 and 5.1 m/s in systole. Infarcted tissue is less contractile following infarction, though not unilaterally stiffer. Single-track-location SWEI (STL-SWEI) is proven to provide suppression of speckle noise and enable improved resolution of structures smaller than 2 mm in diameter compared to ARFI and MTL-SWEI. Contrast to noise ratio and lateral edge resolution are shown to vary with selection of time step for ARFI and arrival time regression filter size for STL-SWEI and MTL-SWEI. </p><p>In 1.5 mm targets, STL-SWEI achieves alternately the tightest resolution (0.3 mm at CNR = 3.5 for a 0.17 mm filter) and highest CNR (8.5 with edge width = 0.7 mm for a 0.66 mm filter) of the modalities, followed by ARFI and then MTL-SWEI.</p><p>In larger, 6 mm targets, the CNR-resolution tradeoff curves for ARFI and STL-SWEI overlap for ARFI time steps up to 0.5 ms and kernels \leq1 mm for STL-SWEI. STL-SWEI can operate either with a 25 dB improvement over MTL-SWEI in CNR at the same resolution, or with edge widths 5×\times as narrow at equivalent CNR values, depending on the selection of regression filter size. Ex vivo ablations are used to demonstrate that ARFI, STL-SWEI and MTL-SWEI each resolve ablation lesions between 0.5 and 1 cm in diameter and gaps between lesions smaller than 5 mm in 3-D scans. Differences in contrast, noise, and resolution between the modalities are discussed. All three modalities are also shown to resolve ``x''-shaped ablations up to 22 mm in depth with good visual fidelity and correspondence to surface photographs, with STL-SWEI providing the highest quality images. Series of each type of image, registered using 3-D data from an electroanatomical mapping system, are used to build volumes that show ablations in in vivo canine atria. In vivo images are shown to be subject to increased noise due to tissue and transducer motion, and the challenges facing the proposed system are discussed. Ultimately, intracardiac acoustic radiation force methods are demonstrated to be promising tools for characterizing dynamic myocardial elasticity and imaging radiofrequency ablation lesions.</p>Dissertatio

    Rock and stiff-Soil site amplification: Dependency on VS30 and Kappa (κ0)

    A ground‐motion prediction equation (GMPE) specific to rock and stiff‐soil sites is derived using seismic motion recorded on high VS30 sites in Japan. This GMPE applies to events with 4.5≤Mw≤6.9 and VS30 ranging from 500 to 1500  m/s (stiff‐soil to rock sites). The empirical site coefficients obtained and the comparison with the simulated site functions show that seismic motion on rock and stiff‐soil sites does not depend only on VS30, but also on the high‐frequency attenuation site properties (κ0). The effects of the site‐specific κ0 on site amplification are analyzed using stochastic simulations, with the need to take into account both of these parameters for rock‐site adjustments. Adding the site‐specific κ0 into the GMPEs thus appears to be essential in future work. The rock‐site stochastic ground‐motion simulations show that the site‐specific κ0 controls the frequency corresponding to the maximum response spectral acceleration (famp1). This observation is used to link the peak of the response spectral shape to κ0 in this specific Japanese dataset and then to add the effects of high‐frequency attenuation into the previous GMPE from the peak ground acceleration and up to periods of 0.2 s. The inclusion of κ0 allows the observed bias to be corrected for the intraevent residuals and thus reduces sigma. However, this κ0 determination is limited to a minimum number of rock‐site records with Mw≥4.5 and to distances of less than 50 km

    Good vibrations: A novel method for sexing turtles

    The ability to accurately determine the sex of individuals is important for research and conservation efforts. While most species of turtle exhibit secondary sexual dimorphisms that can be used to reliably infer sex, there are some species that are very difficult to sex, and even within many dimorphic species, it is not uncommon to encounter individuals that appear to exhibit both male and female secondary sex characteristics. Therefore, we tested the novel method of using a vibrator to sex turtles by stimulating male turtles to evert their penises. We tested this method on males of four species (three families) with known sexual dimorphisms: spiny softshell turtles (Apalone spinifera; n = 14), western chicken turtles (Deirochelys reticularia miaria; n = 17), Mississippi mud turtles (Kinosternon subrubrum hippocrepis; n = 10), and common musk turtles (Sternotherus odoratus; n = 9). The method accurately sexed 100% of A. spinifera, 64.7% of D. r. miaria, 80.0% of K. s. hippocrepis, and 55.6% of S. odoratus. Despite the low success rates in some species, there are situations in which this method will be useful for researchers working with species that are difficult to sex using external morphological characteristics

    Large-scale sedimentation on the glacier-influenced polar North Atlantic Margins: Long-range side-scan sonar evidence

    Long-range side-scan sonar (GLORIA) imagery of over 600,000 km² of the Polar North Atlantic provides a large-scale view of sedimentation patterns on this glacier-influenced continental margin. High-latitude margins are influenced strongly by glacial history and ice dynamics and, linked to this, the rate of sediment supply. Extensive glacial fans (up to 350,000 km³) were built up from stacked series of large debris flows transferring sediment down the continental slope. The fans were linked with high debris inputs from Quaternary glaciers at the mouths of cross-shelf troughs and deep fjords. Where ice was slower-moving, but still extended to the shelf break, large-scale slide deposits are observed. Where ice failed to cross the continental shelf during full glacials, the continental slope was sediment starved and submarine channels and smaller slides developed. A simple model for large-scale sedimentation on the glaciated continental margins of the Polar North Atlantic is presented

    The development of post-processing algorithm for the ultrasonic evaluation by the application of automated robotic testing systems

    The implementation of automated testing systems based on six degrees of freedom (DOF) robotic manipulators is the actual trend in ultrasonic testing equipment development. Such systems are able to provide a fast ultrasonic evaluation with the respect of the surface of the testing object. In this work, the post-processing algorithm based on Synthetic Aperture Focusing Technique (SAFT) is suggested. Such algorithm allows presenting the results in the form of high-resolution imagery of the internal structure of testing objects. The suggested algorithm is applicable in the case of the utilization of automated testing systems based on six DOF robotic manipulators and takes into account all the features conditioned by such equipment application.     Supporting non-target identification by adding hydrogen deuterium exchange MS/MS capabilities to MetFrag

    Liquid chromatography coupled with high-resolution mass spectrometry (LC-HRMS) is increasingly popular for the non-targeted exploration of complex samples, where tandem mass spectrometry (MS/MS) is used to characterize the structure of unknown compounds. However, mass spectra do not always contain sufficient information to unequivocally identify the correct structure. This study investigated how much additional information can be gained using hydrogen deuterium exchange (HDX) experiments. The exchange of “easily exchangeable” hydrogen atoms (connected to heteroatoms), with predominantly [M+D]+ ions in positive mode and [M-D]− in negative mode was observed. To enable high-throughput processing, new scoring terms were incorporated into the in silico fragmenter MetFrag. These were initially developed on small datasets and then tested on 762 compounds of environmental interest. Pairs of spectra (normal and deuterated) were found for 593 of these substances (506 positive mode, 155 negative mode spectra). The new scoring terms resulted in 29 additional correct identifications (78 vs 49) for positive mode and an increase in top 10 rankings from 80 to 106 in negative mode. Compounds with dual functionality (polar head group, long apolar tail) exhibited dramatic retention time (RT) shifts of up to several minutes, compared with an average 0.04 min RT shift. For a smaller dataset of 80 metabolites, top 10 rankings improved from 13 to 24 (positive mode, 57 spectra) and from 14 to 31 (negative mode, 63 spectra) when including HDX information. The results of standard measurements were confirmed using targets and tentatively identified surfactant species in an environmental sample collected from the river Danube near Novi Sad (Serbia). The changes to MetFrag have been integrated into the command line version available at http://c-ruttkies.github.io/MetFrag and all resulting spectra and compounds are available in online resources and in the Electronic Supplementary Material (ESM)
